I think they are just a waste of money for marketing value. Protecting your brand name, I get. I have used two-letter domains, but only as a shortner.

Domains are never all gone. It just takes a little brainstorming, and luck.

The big winners are going to be domain registrars. They often sell those vanity TLD for a premium. Remember when .xxx was $100+ a year? Crazy! Only time will tell if these new domains will get any traction.
